PCR protocols
2µl of each template DNA
total reaction volume of 25 ul
0.15 µM dinucleotide triphosphates (dNTPs)
2.5 mM MgCl2,
0.75 units of Taq polymerase
0.6 uM each primer
PCR conditions
initial 2min at 95C (denaturation)
1 min at 95C (denaturation)
52C for 1 min (primer annealing)
1 min at 72C (amplification) for 30 cycles
5 min at 72C
PCR products are fractionated on 0.9% agarose gels, and bands were visualized by ethidium bromide staining and viewed with ultraviolet light
